Διαδέχτηκε τον γαμπρό του Αλέξιο Α΄, τον ιδρυτή της αυτοκρατορίας, ως κηδεμόνας του νεαρού γιου του Ιωάννη Α΄ Μεγάλου Κομνηνού. Στα τελευταία χρόνια της βασιλείας του Ανδρονίκου Α΄ οι Τραπεζούντιοι συμμάχησαν με τους Μογγόλους της Περσίας, αλλά όταν οι τελευταίοι ηττήθηκαν από τους Σελτζούκους, ο Ανδρόνικος Α΄ έγινε πάλι φόρου υποτελής των σουλτάνων του Ικονίου.
