Officiellement présentée le lundi 9 février 2009, lors d'une séance d'essais sur le circuit de Jerez, cette monoplace tient compte des modifications de règlements introduites en 2009 et présente un aileron avant élargi, un aileron arrière rétréci et relevé et des pneus slicks. Ses pilotes sont l'Australien Mark Webber et l'Allemand Sebastian Vettel, transfuge de l'écurie B Toro Rosso. La RB5 effectue son premier roulage le lundi 9 février 2009. À cause de problèmes techniques lors du développement du SREC, l'écurie a acheté la version développée par Renault mais ne l'a installée dans aucune de ses monoplaces. Christian Horner s'est dit peu séduit par le système de récupération d'énergie cinétique : « En termes de gains de performance, nous ne sentons pas qu'il mérite sa place sur notre voiture, mais cela ne veut pas dire que nous ne l'utiliserons pas à l'avenir [...] À l'heure actuelle, il n'y a pas de date prévue quant à son utilisation ». The car gave the team its first pole position, first win and first ever 1–2 finish at the 2009 Chinese Grand Prix. Over the course of the season the car turned out to be competitive as it won 6 out of 17 races, with Vettel winning four races and Webber winning two. As a result, the team finished 2nd in the Constructors' Championship standings behind Brawn GP and Vettel finished second in the Drivers' Championship standings behind Jenson Button. In July 2010, Red Bull gifted designer Adrian Newey a complete RB5 car as a "thank you" gift for turning Red Bull into a title-challenging team. Het team won 6 van de 17 races met deze wagen: 4 ervan werden behaald door Sebastian Vettel, 2 door Mark Webber. Daarmee werd het team tweede in het constructeurskampioenschap, na Brawn GP. Sebastian Vettel, die sinds hij bij Toro Rosso reed de gewoonte had zijn wagen een naam te geven, noemde de RB5 "Kate".
