Gore hrál univerzitní fotbal za , poté byl vybrán ve třetím kole Draftu NFL 2005 týmem San Francisco 49ers, za který hrál až do roku 2014. V tomto klubu je držitelem rekordu v počtu naběhaných yardů a běhových touchdownů.
