- Denufosol (-) (INS37217), dosud[kdy?] neschválený lék k inhalaci, je antagonistou P2Y2 receptorů, aktivuje epitelových buněk plic, takže je alespoň částečně korigován transport vody a solí mukoviscidózou poškozenými CTFR-kanály. Díky tomu je normalizován dehydratovaný kapalinový film na povrchu epitelových buněk, frekvence kmitů cilií je zvyšována a mukociliární clearance (odstranění hlenu ciliemi) se zlepšuje. Denufosol by měl být využíván obzvlášť při léčbě časného postižení plic. (cs)
- Der noch nicht zugelassene Wirkstoff Denufosol (-Tetranatrium) (INS37217) ist ein P2Y2-Rezeptoren-Agonist zur Inhalation, der in den Epithelzellen der Lunge einen alternativen Chloridkanal aktiviert, so dass der bei der Krankheit Mukoviszidose durch den defekten CFTR-Kanal gestörte Salz- und Wassertransport zumindest teilweise korrigiert wird. Dadurch wird der dehydrierte Flüssigkeitsfilm normalisiert, die Zilienschlagfrequenz erhöht sich, und die Mukoziliäre Clearance (der Schleimabtransport durch die Zilien) wird verbessert.Denufosol soll insbesondere im Frühstadium der Lungenbeteiligung den Verlauf günstig beeinflussen.Es wurden mehrere Phase-3-Studien durchgeführt, die zum Teil gute, zum Teil keine statistisch signifikanten Ergebnisse erbrachten.Die Entwicklung wurde von der amerikanischen Cystic Fibrosis Foundation gefördert.Untersuchungen desselben Stoffes zur Behandlung von Erkrankungen der Retina wurden 2006 aufgegeben. - Denufosol (INN) is an inhaled drug for the treatment of cystic fibrosis, being developed by and sponsored by the Cystic Fibrosis Foundation. It was tested in two Phase III clinical trials, TIGER-1 and TIGER-2. Initially, in the first Phase III trial, TIGER-1, the compound showed significant results as compared with placebo. In the second Phase III trial, TIGER-2, the compound did not meet the primary endpoint, a significant change in baseline FEV1 (forced expiratory volume in one second) at the week 48 endpoint as compared to placebo. As of 2011, no additional clinical studies are being conducted with the compound. The drug was also investigated for the treatment of retinal detachment and other retinal diseases, but trials were terminated in 2006. (en)
