It was also used off-label by individuals who wished to avoid fatigue, such as night workers or others who needed to stay awake and alert for long periods of time. Additionally, the medication has been used non-medically as a nootropic. Adrafinil is a prodrug; it is primarily metabolized in vivo to modafinil, resulting in very similar pharmacological effects. Unlike modafinil, however, it takes time for the metabolite to accumulate to active levels in the bloodstream. Effects usually are apparent within 45–60 minutes when taken orally on an empty stomach. Adrafinil was marketed in France until September 2011 when it was voluntarily discontinued due to an unfavorable risk–benefit ratio. (en)
- Adrafinil ist ein Arzneistoff zur Behandlung von Schlafstörungen wie z. B. der Narkolepsie. Die Substanz gehört zu einer Gruppe psychostimulierender Medikamente, die sich in ihrer Molekülstruktur von den Stimulanzien auf Amphetaminbasis deutlich unterscheidet. Adrafinil ist ein Prodrug, das erst durch Metabolisierung zu Modafinil wirksam wird. In seiner Wirkung entspricht es Modafinil, jedoch setzt aufgrund des Metabolisierungsschritts die Wirkung verzögert ein. Es ist wie Modafinil eine verbotene Dopingsubstanz. Adrafinil wurde in Frankreich ab 1985 unter dem Produktnamen Olmifon® vertrieben. Nachdem die französische Arzneimittelbehörde dem Hersteller Cephalon im April 2011 angekündigt hatte, dass eine Neubewertung des Nutzen-Risiko-Verhältnisses von Olmifon geplant sei, stellte Cephalon den Vertrieb ein. También se utiliza fuera de indicación para evitar la fatiga y la somnolencia, como por ejemplo trabajadores nocturnos y aquellos que necesitan mantenerse alerta y despiertos durante largos períodos de tiempo. El Adrafinil es un profármaco, principalmente metabolizado in vivo a modafinilo, dando como resultado efectos farmacológicos casi idénticos. A diferencia del modafinilo, transcurre cierto tiempo hasta que el metabolito se acumula a niveles activos en el torrente sanguíneo. Los efectos generalmente son evidentes dentro de 45-60 minutos cuando se toma por vía oral con el estómago vacío. Adrafinil no tiene actualmente la aprobación de la FDA y por lo tanto no está reglamentado en Estados Unidos. Se comercializó en Francia y otros países de Europa bajo el nombre comercial de Olmifon hasta septiembre de 2011, cuando el equivalente francés de la FDA revaluó el fármaco y retiró el permiso de comercialización. Il farmaco è stato usato per alleviare l'eccessiva sonnolenza e disattenzione nei pazienti anziani. È stato inoltre utilizzato off-label da parte di individui che desiderano evitare la fatica, ad esempio i lavoratori notturni o altri soggetti che necessitano di restare svegli e vigili per lunghi periodi di tempo. Adrafinil è un profarmaco, che viene metabolizzato in vivo in modafinil, con effetti farmacologici quasi identici a quest'ultimo composto. Adrafinil non ha ricevuto l'autorizzazione al commercio da parte della USA Food and Drug Administration (FDA) ma è stato commercializzato in Francia e in altri Paesi europei con il nome commerciale di Olmifon fino al dicembre 2011 quando l'Agence nationale de sécurité du médicament et des produits de santé (ANSM) ha rivalutato il farmaco concludendo che il rapporto rischio/beneficio dell'adrafinil è sfavorevole e proponendone quindi una revoca dell'autorizzazione al commercio.Un lavoro scientifico del 2007 ha messo in luce come il farmaco possa essere oggetto di utilizzo in ambito sportivo o militare come psicostimolante, anche in un contesto di automedicazione. - Adrafinil – organiczny związek chemiczny będący prolekiem, który w wątrobie metabolizowany jest do biologicznie aktywnego modafinilu o wyraźnym działaniu podtrzymującym stan czuwania i redukującym uczucie senności (co pozwala zaliczyć go do tzw. nootropów, lub ściślej do ) oraz stosunkowo łagodnym działaniu stymulującym, a także o pewnym niewielkim potencjale przeciwdepresyjnym.
