Sie prägte die Entwicklung der Konversationsanalyse, zusammen mit Emanuel Schegloff und Harvey Sacks. Von ihr stammt auch das heute vielfach verwendete System zur Transkription gesprochener Sprache, das v. a. in der englischsprachigen Forschung verwendet wird. Dem Vernehmen nach soll Jefferson die ersten Transkriptionen auf Anweisung Sacks' entworfen und vorgenommen haben; er überreichte ihr Bandaufnahmen mit den Worten "Schreib mal auf was du da hörst". She was, along with Harvey Sacks and Emanuel Schegloff, one of the founders of the area of research known as conversation analysis (CA). She is remembered for the methods and notational conventions she developed for transcribing speech, the latter forming the Jefferson Transcription System. This is now used widely in CA research. (en)

Elle a contribué au développement de l'analyse conversationnelle avec Harvey Sacks et Emanuel Schegloff. Sa méthode de transcription des conversations est connue sous le nom de « Jefferson System ». Jefferson est décédé du cancer en mars 2008. Après avoir été chercheur à l'Université de Manchester (Royaume-Uni) (1978-1981), de Tilbourg (Pays-Bas) (1981-1983) et occupé un poste de professeur honoraire à York (1984-1985), elle s'établit aux Pays-Bas .
