In molecular biology, ribosomal s6 kinase (rsk) is a family of protein kinases involved in signal transduction. There are two subfamilies of rsk, p90rsk, also known as MAPK-activated protein kinase-1 (MAPKAP-K1), and p70rsk, also known as S6-H1 Kinase or simply S6 Kinase. There are three variants of p90rsk in humans, rsk 1-3. Rsks are serine/threonine kinases and are activated by the MAPK/ERK pathway. Terdapat dua jenis Rsk, p90rsk atau MAPK-activated protein kinase-1 (MAPKAP-K1) dan p70rsk atau S6-H1 Kinase atau S6K. Manusia memiliki 4 ekspresi Rsk, yaitu RSK-1, RSK-2, RSK-3, RSK-4. Rsk banyak dijumpai pada inti sel, sitoplasma dan membran plasma, terutama pada sel otak dan testis. Pada sistem saraf, Rsk diketahui mempercepat fosforilasi pada protein pro-apoptotik pada serina residu 112, baik in vitro maupun in vivo. Fosforilasi ini mencegah apoptosis neuron yang dimediasi oleh BAD. Katalisasi Rsk juga terjadi pada faktor transkripsi pada serina 133. Fosforilasi CREB berfungsi guna kelangsungan hidup sel, dan terhambatnya fosforilasi ini akan menginduksi apoptosis. RSK-2, ketika teraktivasi, akan bermigrasi menuju ke dalam inti sel dan menkatalis berbagai protein inti seperti , , histon, CREB, dan p53. Pada hewan tikus, terhambatnya RSK-2 meredakan terbentuknya yang bergantung pada c-Fos. Sedangkan senyawa yang merupakan ekstrak tumbuhan tropis dapat menghambat aktivitas RSK-2 dan menekan proliferasi sel kanker payudara dan sel kanker prostat . Mutasi genetik RSK-2 akan menyebabkan penyimpangan pada lintasan MAPK/Rsk hingga menginduksi . (in)
