It was extended to roughly its current length in 1930, replacing NY 76 west of Marcy and utilizing previously unnumbered highways west of Central Square and east of Marcy. The section of NY 49 between Rome and Utica originally followed a surface road alongside the Mohawk River; however, it was rerouted onto the Utica–Rome Expressway as sections of the road opened to traffic.
