La flumequina viene assorbita rapidamente dal tratto gastrointestinale. Dopo somministrazione orale di dosi singole di 400, 800 e 1200 mg si ottengono entro 2 ore livelli plasmatici rispettivamente di 13,5, 23,8 e 31,9 µg/ml, con livelli urinari corrispondenti di 50, 105 e 131 µg/ml misurati dopo 6-8 ore. L'emivita media di eliminazione è di 7,1-8,5 ore. La flumequina subisce metabolismo epatico e viene escreta nelle urine sotto forma di glucuronidi inattivi e di un idrossi-derivato attivo; solo una piccola quota viene escreta in forma immodificata. Dopo somministrazione orale la DL50 nel topo e nel ratto è rispettivamente di 1230 e di 776 mg/kg. La flumequina è impiegata nel trattamento di infezioni delle vie urinarie sostenute da microorganismi sensibili. Inoltre, è indicata nella prevenzione e nel trattamento di infezioni che possono insorgere nel corso di interventi chirurgici, litiasi biliare o manovre endoscopiche. Si somministrano 400 mg 2-3 volte al giorno, per via orale durante i pasti. La terapia deve essere protratta per almeno dieci giorni. La flumequina può causare nausea, vomito, diarrea, gastralgie, aumento della peristalsi, rash cutanei, prurito, fotosensibilizzazione, turbe dell'accomodamento, stordimento transitorio, vertigini. La flumequina è controindicata in età pediatrica e in caso di ipersensibilità accertata al farmaco o ai chinolonici in generale. Si somministra con estrema cautela in gravidanza e nelle gravi forme di insufficienza renale o epatica. Durante la terapia con flumequina si consiglia di evitare l'esposizione ai raggi solari. La flumequina può antagonizzare l'azione di tetracicline e nitrofurantoina. Si osserva sinergismo con gentamicina e colistina. (it)
