- La chromatographie bidimensionnelle (ou deux dimensions) résulte du couplage de deux séparations chromatographiques de nature différente dans le but de séparer des mélanges complexes de substances similaires. Le principe est de découper le premier chromatogramme en plages de quelques secondes (cinq à dix). Tous les composés élués durant chacune de ces plages sont ensuite soumis à une nouvelle chromatographie dont les critères de séparation diffèrent de la première. Bien que le principe de cette technique remonte à plusieurs années[Quand ?], les moyens informatiques (principalement la puissance de traitement des ordinateurs) permettant l'analyse des résultats sont récents[Quand ?]. Cette procédure peut être appliquée à la chromatographie sur colonne (regroupant notamment l'HPLC et la CPG) et à la chromatographie planaire (qui recouvre la CCM et la chromatographie sur papier). Dans le cas de la chromatographie planaire, celle-ci s'effectue en deux étapes entre lesquelles on change de solvant et on tourne le papier de 90°. Les interactions développées par le nouveau solvant seront différentes, ce qui modifiera la séparation dans cette deuxième dimension et permettra une meilleure séparation globale. (fr)
- Two-dimensional chromatography is a type of chromatographic technique in which the injected sample is separated by passing through two different separation stages. Two different chromatographic columns are connected in sequence, and the effluent from the first system is transferred onto the second column. Typically the second column has a different separation mechanism, so that bands that are poorly resolved from the first column may be completely separated in the second column. (For instance, a C18 reversed-phase chromatography column may be followed by a phenyl column.) Alternately, the two columns might run at different temperatures. During the second stage of separation the rate at which the separation occurs must be faster than the first stage, since there is still only a single detector.
