- N-Glycolylneuraminsäure, auch Neu5Gc genannt, gehört zu der Klasse der Sialinsäuren. In tierischen Organismen ist Neu5Gc eine häufig vorkommenden Sialinsäure. Im menschlichen Organismus dagegen fehlt Neu5Gc. Beim Menschen kommt ausschließlich N-Acetylneuraminsäure vor, da das Enzym CMP-Neu5Ac-Hydroxylase, welches die Bildung von N-Glycolylneuraminsäure aus N-Acetylneuraminsäure katalysiert, beim Menschen nicht funktional ist. Das Gen, welches für das entsprechende Enzym (CMP-Neu5Ac-Hydroxylase = ) codiert, ist beim Menschen durch eine Mutation funktionslos. Im Gegensatz dazu haben beispielsweise Schimpansen die Fähigkeit Neu5Gc – unter anderem im Gehirn – zu synthetisieren. Das für CMAH codierende Gen befindet sich auf Chromosom 6 Genlocus p21.32. - N-Glycolylneuraminic acid (Neu5Gc) is a sialic acid molecule found in most non-human mammals. Humans cannot synthesize Neu5Gc because the human gene CMAH is irreversibly mutated, though it is found in other apes. It is absent in human tissues because of inactivation of gene encoding CMP-N-acetylneuraminic acid hydroxylase. The gene CMAH encodes for CMP-N-acetylneuraminic acid hydroxylase, which is the enzyme responsible for CMP-Neu5Gc from CMP-N-acetylneuraminic (CMP-Neu5Ac) acid. This loss of CMAH is estimated to have occurred 2-3 million years ago, just before the emergence of the genus Homo. (en)
