- Circular molecules of DNA, such as plasmids and typical mitochondrial genomes, consist of two strands of DNA called the heavy strand (or H-strand) and the light strand (or L-strand). The two strands have different masses due to different proportions of heavier nucleotides. While this difference is not known to have any functional significance, it can be used in the laboratory to segregate the strands of denatured DNA, and hence to analyze the strands separately. Adenine and guanine (purines) are heavier than cytosine and thymine (pyrimidines) due to their extra ring. Because a purine always pairs with a pyrimidine, any excess of purines in one strand will occur with a corresponding excess of pyrimidines in the other strand and vice versa. Statistically, there is more likely to be such an imbalance than an exact 50/50 ratio. In addition, bias may arise due to differentials in the amount of protein-coding sequence on each strand, as codons do not all occur with equal frequency. (en)
- Les molécules circulaires d’ADN, telles que les plasmides et les génomes typiques des mitochondries, sont constituées de deux brins d’ADN appelés respectivement brin lourd (H-strand pour Heavy strand en anglais) et brin léger (L-strand pour Light strand en anglais). Les deux brins ont des masses différentes en raison des proportions différentes en nucléotides lourds. Bien que l’on n’ait pas connaissance de différences fonctionnelles qui y soient associées, cette propriété peut être mise à profit en laboratoire car elle permet de séparer les brins d’ADN après dénaturation et ainsi de les analyser séparément. Parce qu’elles possèdent un cycle supplémentaire, les bases azotées purines (adénine et guanine) de l’ADN sont plus lourdes que les bases azotées pyrimidines (thymine et cytosine). Comme une purine d’un brin s’associe toujours à une pyrimidine sur l’autre brin, un excès de purines sur un brin sera toujours lié à un excès de pyrimidines sur l’autre brin, et vice versa. (fr)
