Wittenberg war Hauptstadt des sächsischen Kurkreises und einst herzogliche und kurfürstliche Residenz von Sachsen-Wittenberg. Od roku 1502 je univerzitním městem. Wittenberk se stal po příchodu Martina Luthera v roce 1508 a uveřejněním jeho 95 tezí v roce 1517 kolébkou reformace a evangelické teologie. Působily zde další významné osobnosti, spojené s reformací, mj. Philipp Melanchthon a malíř Lucas Cranach. Na zdejší univerzitě studovali i čeští učenci, např. Jan Blahoslav nebo Jiří Třanovský. V zámeckém kostele, ve kterém kázal, je pochován Martin Luther, i jeho poradce Philipp Melanchthon. Od roku 1996 je několik budov ve městě zapsáno na Seznamu světového kulturního dědictví UNESCO.

Resultó seriamente dañada por un incendio en 1760 a consecuencia de un bombardeo francés durante la Guerra de los Siete Años, y fue prácticamente reconstruida y restaurada años más tarde. Las puertas de madera, quemadas en 1760, fueron sustituidas en 1858 por las puertas de bronce, llevando el texto latino de las tesis. Dentro de la iglesia están las tumbas de Lutero y de Philipp Melanchthon, de los electores Federico el Sabio (por Peter Vischer, 1527) y Juan la constante, y los retratos de los reformadores de Lucas Cranach el Joven.
