Mehmet 1648 yılında 6 yaşındayken tahta geçince Valide Sultan oldu. Ancak kayınvalidesi eski Valide Sultan Kösem Sultan halen hayattaydı ve devlet işlerinde etkili olmaya devam ediyordu. Turhan Sultan'la Kösem Sultan arasında kıyasıya bir rekabet başladı. Bu rekabet 3 yıl sürdü ve Kösem Sultan'ın bir gece dairesinin basılarak boğdurulmasıyla noktalandı. Turhan Sultan toplam 34 yıl Valide Sultanlık yaparak Osmanlı tarihinin en uzun süreli Valide Sultan'ı oldu. Bu dönemde Köprülü ailesinden sadrazamlar iş başına geldi ve Valide Sultanların devlet siyasetindeki etkileri azaldı. Turhan Sultan 1597'de Sultan III. Mehmet'in annesi Valide Safiye Sultan'ın emriyle yapımına başlanan fakat yarım kalan İstanbul Yeni Camiini kendi parasıyla tamamlattı. 1683 yılında öldü ve cenazesi kendi tamamlattığı Yeni Camii'nin avlusundaki Turhan Sultan Türbesine gömüldü.
