Sein Broadway-Debüt hatte er 1941 in dem Musical Pal Joey von Richard Rodgers und Lorenz Hart. Nach dem Zweiten Weltkrieg holten ihn der Filmproduzent Arthur Freed und der Choreograf Gene Kelly nach Hollywood, um das Musical On the Town von Leonard Bernstein zu verfilmen. Damit begann seine Karriere als er der wichtigste Musicalregisseur von MGM in den 1950er Jahren, in denen er unter anderem den Klassiker der MGM-Musicals, Singin' In The Rain (1952), inszenierte. Allerdings erhielt Donen nie einen Oscar für seine Arbeiten. Erst 1998 wurde ihm der Oscar für sein Lebenswerk verliehen. 2004 erhielt er bei den Filmfestspielen von Venedig den Ehrenlöwen.
