Smallpox vaccine, the first successful vaccine to be developed, was introduced by Edward Jenner in 1796. Il termine vaccino deriva dalla parola variolae vaccinae (cioè vaiolo della mucca), il termine ideato da Jenner per indicare vaiolo bovino. Il termine vaccinazione sostituì presto la dizione inoculazione da vaiolo della mucca, e fu usata per la prima volta in un documento che fu dato alle stampe da un amico di Jenner, Richard Dunning nel 1800. O desenvolvimento da técnica resultou da observação feita por Jenner que as raparigas que vendiam leite de vaca não eram atacadas pela varíola desde que tivessem sido anteriormente infectadas pela varíola bovina, o que mostrava que a a inoculação com varíola bovina, bem mais benigna, protegia contra a varíola. A palavra "vacina" é derivada de Variolae vaccinae, a varíola dos bovinos, o termo cunhado por Jenner para designar a varíola bovina e usado no título da sua obra An enquiry into the causes and effects of Variolae vaccinae, known by the name of cow pox.
