Romain aimait les offices de l'Église mais regrettait de ne pouvoir bien les chanter à cause de sa voix peu agréable. Un jour qu'il assistait à la vigile de Noël dans l'église des Blachernes, la Mère de Dieu lui apparut tenant un parchemin roulé qu'elle lui donna à manger. Aussitôt qu'il eut goûté cette nourriture particulière, il se mit à chanter à pleine voix un kontakion que l'on chante toujours la nuit de Noël : "La Vierge aujourd'hui met au monde l'éternel... " Romain le Mélode passe pour avoir composé plus de mille kontakia parmi lesquels on compte le fameux Acathiste à la Mère de Dieu.

Diacono, in seguito coadiutore nella chiesa della Resurrezione a Berito, quando si recò a Costantinopoli, allora sotto l'imperatore Anastasio, si ritirò nella chiesa della Madre di Dio. Morì circa nel 555. Di lui ha detto papa Benedetto XVI: "Appartiene alla grande schiera dei teologi che hanno trasformato la teologia in poesia", come sant'Efrem di Siria, sant'Ambrogio, san Tommaso d'Aquino, san Giovanni della Croce e tanti altri.
