Deze synthesizer was populair, klonk goed en was eenvoudig in het gebruik. Men kon er snel en intuïtief geluiden mee samenstellen, via een overzichtelijke set schuiven en (draai)knoppen. Er pasten 128 patches in het geheugen, wat erg veel was voor die tijd. De Juno-106 valt op vanwege het feit dat hij een mooie sound heeft terwijl er slechts één oscillator per stem wordt gebruikt. De oscillator kon twee golfvormen bij elkaar mixen. terwijl het toen al gebruikelijk was minstens twee oscillatoren per stem aan te wenden, hetgeen het instrument veelzijdiger maakt. Dat de Juno-106 toch vol klinkt is mede te danken aan de goede kwaliteit van het ingebouwde chorus. De Juno-106 had een hoog-kwalitatieve en voor die tijd zeer volledige MIDI-implementatie: de 16 MIDI-kanalen werden allemaal gebruikt en ook SysEx data kon worden verstuurd en ontvangen, evenals controllerdata voor alle schuiven. Daardoor werkte de Juno naadloos samen met sequencers. De Juno-106 klinkt het best bij strings en pad-geluiden. Gefilterde sweeps klinken ook goed, en de bassen zijn mooi en rond van klank. Solo-geluiden ermee maken lukt niet echt goed, men ervaart dan ernstig het gemis van de tweede oscillator. Het geluid blijft te dun. Ook voor punchy en funky akkoordbegeleidingen heeft de Juno-106 net iets te weinig in huis. De juno-106 klinkt minder 'vet' dan zijn voorganger, de Juno-60 door de gebruikte componenten en aansturing. Qua klank is de Juno-106 te vergelijken met de Roland JX-3P, alleen heeft deze meer functies waaronder een tweede oscillator, crossmodulatie en een simpele sequencer.
