Es handelt sich dabei um eine Abhandlung über das Lebensgefühl Melancholie, ihre Geschichte, Ursachen und Heilmöglichkeiten. In diesem Buch prägte Burton auch das bekannte Gleichnis Auf den Schultern von Giganten, dessen eigentlicher Urheber jedoch Bernhard von Chartres ist (siehe auch die Fenster in der Kathedrale von Chartres), und begründete erste Vorläufer der Kognitionswissenschaft.

La sua prosa, costellata di citazioni in latino e in greco, ha un carattere bizzarro. Nonostante il suo senso dell'umorismo, Burton non riesce a nascondere l'inquietudine che suscita in lui la patologia depressiva. La prima edizione inglese aveva 900 pagine, e conteneva 350 mila parole; l'ultima era lunga 1500 pagine e conteneva più di 500 mila parole. Nel testo sono state contate 13333 citazioni di 1598 autori differenti. Burton è inoltre l'autore di Philosophaster, una commedia di carattere satirico in lingua latína.
