Nicolas Cotoner, Grand Master of the Order of between 1663-1680, continued the work on the redecoration of St. John’s Co-Cathedral commenced by his brother, Rafael, and much of the splendour of the Cathedral occurred during his reign including the decoration of the vault painted by the Calabrian artist Mattia Preti as well as much of the carving and gilding of most of the walls. The sepulchral monument to Grand Master Nicolas Cotoner, located prominently to the right side of the main altar in the chapel of the langue of Aragon in St John's Co-Cathedral, was produced by Domenico Guidi and is one of the most prominent and beautiful monuments in the Cathedral. The remarkable Cotoner monument consists of a pyramidal distribution of figures with a central grouping of triumphal paraphernalia such as arms and trophies which surround the bronze gilded bust of the Grand Master. Above a cherub holds the Cotoner armorial shield whilst the allegory of Fame blows a trumpet in triumph. The sepulchral monument was assembled in the chapel in June of 1686.

Während seiner Regierungszeit war der Künstler Mattia Preti in St. Johann tätig, der damaligen Hauptkirche des Ordens und heutigen St. John's Co-Kathedrale in Valletta, in der Nicolas Cotoner auch seine letzte Ruhestätte fand. Nicolas Cotoner war hartnäckig, ungestümen und schnell erregbar. Er hatte nicht nur an Pretis Arbeiten großes Interesse, sondern auch an der Verbesserung der Sacra Infermeria, dem größten Ordenshospital. 1676 gründete er hier eine medizinische Hochschule, die Schule für Anatomie und Chirurgie. Ebenso entstanden zu dieser Zeit riesige Festungsanlagen, die Cottonera Lines. Beide Cotoner waren große Baumeister und ihr Wappen ist in den meisten Fassaden der Paläste, Kirchen und Festungen in Valletta und in anderen Teilen der Insel eingemeisselt. Allerdings bleibt St. Johann ihr größter Beitrag, den sie beide liebten und verschönerten. Es war eine Zeit des Wohlstands auf den Inseln, aber ihre Manie für den Aufbau und ihr extravaganter Geschmack waren eine Belastung für die Staatskasse.
  • Nicolau Cotoner i d'Olesa fou Gran Mestre de l'Orde de Malta, entre 1663 i 1680. Era fill de Marc Antoni Cotoner i de Santmartí i germà de l'anterior Mestre. Va ingressar a l'orde el 1625 i va anar a fer el noviciat a Malta al costat del seu germà Rafael. El 1644 fou nomenat castellà de Malta i el 1645 batlle de Negropont. El 1660 va succeir el seu germà primer a la batllia de Mallorca i el 1663 en el càrrec de Gran Mestre. En el seu mestratge va fer construir unes defenses per a la població civil a Malta, anomenada la Cotonera, ja que Venècia i el Turc havien firmat la pau i temia represàlies d'aquests últims. Es va dedicar a l'estratègia i a la diplomàcia i així l'Orde va créixer en prestigi a diferents estats europeus, com França o la Gran Bretanya. El 1674 va fundar una escola d'anatomia anexa a la Infermeria de l'Orde que el seu germà havia fet ampliar i aquest mateix any va redactar en italià les constitucions i estatuts de l'Orde.

Prima di essere eletto Gran Maestro era stato Priore d'Aragona e come tale, alla sua morte, venne sepolto nella Cappella d'Aragona della cattedrale dell'Ordine. Gli succedette Fra Gregorio Carafa.
