He was a member of the North Carolina House of Commons in 1820, serving as Speaker of the House, and was a member of the North Carolina Senate from 1823 to 1827. He was elected to the United States House of Representatives in 1828, serving from 1829 to 1837, not being a candidate for renomination in 1836.

Speight died in Columbus, Mississippi on May 1, 1847 and was interned in Friendship Cemetery in Columbus.

Auch dort blieb er politisch aktiv und wurde 1841 in den Landessenat gewählt, in dem er bis 1844 verblieb. Von 1842 bis 1843 war er dabei Vizegouverneur von Mississippi und fungierte somit als Präsident des Senats. Schließlich kehrte er nach erfolgreicher Wahl im Jahr 1845 als US-Senator in den Kongress zurück, wo er den Vorsitz zweier Ausschüsse innehatte. Jesse Speight starb jedoch noch während seiner Amtszeit im Mai 1847 in Columbus, wo er auch beigesetzt wurde.
