Partecipò a quattro edizioni del Giro d'Italia tra il 1932 ed il 1935, classificandosi al secondo posto nel 1932 e nel 1933. Nelle corse in linea colse il successo più importante alla Milano-Sanremo del 1934. Fu anche campione nazionale di ciclocross nel 1932.

In 1928 startte Joseph, in de volksmond “Tjeppen Demuysere” als profrenner bij het team Lucifer-Hutchinson, waar hij direct hoge ogen gooide. Het hoogtepunt van zijn carrière vormt zijn overwinning in Milaan-San Remo in 1934, maar hij kan ook teren op een aanzienlijk palmares in de Tour de France : 3de in 1929 en 2de in 1931. In de Ronde van Italië eindigde hij 2de in 1932 en 3de in 1933. In 2007 staat Joseph Demuysere in het daglicht in zijn geboorteplaats Wervik, waar in dit Joseph-Demuyserejaar verschillende manifestaties gepland staan, een borstbeeld werd onthuld, en een straat naar deze verdienstelijke wielrenner werd genoemd.
