James Semple was a United States Senator from Illinois. Born in Green County, Kentucky, he had some private education as well as public schooling before enlisting in the Army in 1814 and being an ensign in the Kentucky Militia in 1816. He moved to Edwardsville, Illinois, in 1818 and to Chariton, Missouri, in 1819. Semple unsuccessfully ran for U.S. Senate in 1836.

He moved to Edwardsville, Illinois, in 1818 and to Chariton, Missouri, in 1819. Semple unsuccessfully ran for U.S. Senate in 1836. President Martin Van Buren appointed him as Chargé d'Affaires to New Granada on 14 October 1837, and he served in that capacity until 4 March 1842. Semple was the judge of the Illinois Supreme Court from 1842 to 1843. He was appointed and subsequently elected as a Democrat to the United States Senate to fill the vacancy caused by the death of Samuel McRoberts and served from December 4 1843, to March 3, 1847. He was not a candidate for renomination in 1846. Semple ended up in Jersey County, Illinois, where in 1853 he founded the town of Elsah, Illinois. His son Eugene Semple was a politician in the territory and state of Washington. James Semple died in Elsah, Illinois on December 20, 1866 and is buried in the Bellefontaine Cemetery, St. Louis, Missouri.

Von 1828 bis 1833 gehörte er dem Repräsentantenhaus von Illinois an, dessen Speaker er auch zeitweise war. 1832 nahm er am Black-Hawk-Krieg teil, wobei er sowohl als Private an Kampfhandlungen teilnahm als auch dem Militärgericht angehörte. Nach seiner Zeit im Parlament von Illinois wurde er 1833 Attorney General des Staates. Erfolglos verlief 1836 Semples erste Kandidatur für den US-Senat. Wenig später zog er nach Alton, ehe er von Oktober 1837 bis März 1842 das Amt des amerikanischen Chargé d'affaires in der Republik Neugranada bekleidete. Nach seiner Rückkehr war er zwischen 1842 und 1843 Richter am Obersten Gerichtshof von Illinois. Nach dem Tod von US-Senator Samuel McRoberts wurde James Semple zu dessen Nachfolger im Kongress ernannt. Er gewann auch die Nachwahl und gehörte dem Senat damit vom 4. Dezember 1843 bis zum 3. März 1847 an; zur Wiederwahl trat er nicht an. Während dieser Zeit war er Vorsitzender des Committee on Revolutionary Claims. In der Folge lebte Semple zunächst wieder als Geschäftsmann in Alton, ehe er 1853 ins Jersey County zog, wo er den Ort Elsah gründete. Dort starb er dann auch im Dezember 1866.
