Instead, appeals were made to the Portuguese government, which had been driven out of Portugal the year before during the Peninsula War and was resident in Brazil. In exchange for providing troops and transports for the operation, the Portuguese were promised Guiana as an expansion of their holdings in Brazil for the duration of the conflict. The British contribution was small, consisting solely of the minor warship HMS Confiance. Confiance however had a highly effective crew and an experienced captain in James Lucas Yeo, who was to command the entire expedition. The more substantial Portuguese contingent consisted of 700 regular Brazilian Colonial Army soldiers, led by Lieutenant-General Manuel Marques de Sousa, The First and 550 regular soldiers from the naval infantry of the Colonial Navy of Brazil and several warships to act as transports and provide offshore artillery support. The French defenders were weakened by years of Royal Navy blockade and could only muster 400 regular infantry and 800 unreliable militia, formed in part from the territory's free black population. As a result resistance was inconsistent and despite Cayenne's strong fortifications, the territory fell within a week. It is considered to be the baptism of fire of the Brazilian Marine Corps.

Ou seja, a fronteira voltariam ao rio Oiapoque, combinados no Congresso de Utrecht em 1713, anulando o estabelecido em Paris (1797), em Badajoz e em Madrid (1801) e mesmo em Amiens (1802) Por terra foram enviados 700 homens vindos do Pará comandados pelo tenente-general Manuel Marques, enquanto que pelo mar eram apoiados pela esquadra da Marinha Real Britânica, comandada por James Lucas Yeo, chefe das forças navais, a bordo da fragata Confiance com 26 peças de artilharia, enviada do Rio de Janeiro, junto com os brigues Voador (com 18 peças, comandado por José Antônio Salgado) e Infante D. Pedro (com 18 peças comandada por Luís da Cunha Moreira), a escuna General Magalhães com 12 peças e os cúteres Vingança e Leão, comandados pelo tenente Manuel Luís de Melo. Os fuzileiros-marinheiros estavam sob o comando de Luís da Cunha Moreira, mais tarde visconde de Cabo Frio e primeiro ministro da Marinha do Brasil independente . O primeiro combate foi travado em 15 de dezembro, às margens do rio Aproak, duas embarcações francesas são apreendidas. Algumas semanas depois as tropas inglesas e portuguesas partem para conquistar as principais fortificações francesas no rio Maroni. Em 6 de janeiro de 1809 o forte Diamante é conquistado, no dia seguinte o forte Dégrad des Cannes e no outro o forte Trió, ambos na ilha de Caiena. A tropa sitiou a capital, Caiena, cujo governador Victor Hughes se rendeu sem resistência, em 12 de janeiro de 1809, assinando em Bourda a rendição. A Guiana passou a ser administrada por João Severiano Maciel da Costa, depois marquês de Queluz, com a designação de Colônia de Caiena e Guiana.
