The longest-lived incarnation of the act featured Gladys Knight on lead vocals, with The Pips, who included her brother Merald "Bubba" Knight and their cousins Edward Patten and William Guest, as backup singers.

Kurz nach der Single Letter Full of Tears kehrten Langston George und Bubba Knight der Band den Rücken. Als Folge dessen veröffentlichten Gladys Knight & the Pips nur noch wenig eigenes Material, sie halfen nun vor allem anderen Musikern im Studio. In der Besetzung Gladys und Bubba Knight, Edward Patten und William Guest erschien 1964 die Single Giving Up, mit der die Band abermals in höhere Chart-Positionen vorrücken konnte. 1966 nahm Berry Gordy die vier Musiker bei Soul Records unter Vertrag und machte Norman Whitfield zu ihrem Produzenten. Es folgten weitere Hits, von denen I Heard It Through the Grapevine der erfolgreichste wurde. Auch einige Alben der Band kamen nun in die Charts. Ab 1969 wurde Gladys Knight & the Pips von Ashford & Simpson produziert, die Hit-Welle hielt an. Der größte Hit dieser Zeit war Midnight Train to Georgia. Bis 1977 konnten Gladys Knight & the Pips diverse Alben und Songs in den Pop-Charts platzieren. Sie erhielten mehrere Grammys.
