He is esteemed by soul fans as one of the leading exponents of the 1960s Chicago soul scene, along with Curtis Mayfield and Jerry Butler. His signature hit is the #1 Billboard Hot 100 chart song, "Duke of Earl" (1962). Chandler wrote and performed many more songs, and collaborated with many of the greats in the soul and R&B world.

Die meiste Zeit seiner Armeezugehörigkeit verbrachte er in Deutschland. Dann kehrte er zu den Dukays zurück und hatte mit denen 1961 und 1962 zwei kleinere Hits. Nun nahm die Band den Titel Duke of Earl (1962 USA Platz 1) auf, den sie auf einem neuen Plattenlabel veröffentlichen wollten. Ihre alte Plattenfirma verbot ihnen, den Namen Dukays weiter zu verwenden. Bis zu diesem Zeitpunkt nannte sich Gene noch nach seinem Geburtsnamen Eugene Dixon, nunmehr nahm er das Pseudonym Gene Chandler an, veröffentlichte den Song und erreichte damit den Spitzenplatz der Charts. Die Reihe seiner weiteren Hits reicht bis Anfang der 1980er Jahre. Etliche seiner größeren Erfolge wurden von Curtis Mayfield geschrieben, so z.B. Just Be True (1964 USA Platz 19) oder Nothing Can Stop Me (1965 USA Platz 18). Seinen einzigen Millionenseller hatte er mit Groovy Situation (1970 USA Platz 12). 1980 glückte ihm auch ein Sprung in die britische Hitparade. Sein Hit Duke of Earl wurde mehrmals von anderen Gruppen veröffentlicht und war 1991 Grundlage für den Spielfilm King Ralph mit John Goodman.
