Compared by some to Stanley Matthews, Moulijn is considered one of the most talented leftwingers in Dutch football history. Johan Cruyff added him to his alltime favorite Dutch national team, stating that "Coen mastered one movement better than anyone: threatening to pass his opponent through the center, and then speeding past him on the other side. Après s'être fait connaître aux yeux du grand public avec Xerxes, le club de son enfance, Moulijn est contacté par le Feyenoord Rotterdam et signe au club, au grand détriment de son père supporter du Sparta Rotterdam. Le montant du transfert s'élevait alors à 25 000 florins soit 11 350 € aujourd'hui. À Feyenoord, il s’impose rapidement comme un joueur clé de l'équipe grâce a ses qualités technique sur le côté gauche du terrain et est séléctionné en équipe nationale pour la première fois en 1956 pour un match contre la Belgique, il portera 38 fois le maillot Oranje et y inscrira 4 buts. Grâce à ses prestations avec le Feyenoord, il est devenu l'idole des supporters qui lui avaient alors dédié une chanson. Toujours très populaire dans ce club où il passa 17 ans, la mascotte du club a été nommé en son honneur et porte son diminutif, Coentje.

Hij stond bekend om zijn spectaculaire en vaak onnavolgbare acties op de flanken, maar liet het verdedigen over aan zijn vriend Cor Veldhoen. In later jaren was hij samen met Ove Kindvall de publiekslieveling bij de voetbalclub uit Rotterdam. In 1970 won hij met Feyenoord de finale van de Europa Cup I ten koste van het Schotse Celtic. In totaal kwam Moulijn 487 keer uit in het eerste elftal van Feyenoord, waarmee hij de clublijst aanvoert. Nog tijdens zijn actieve voetbalcarrière begon hij in 1961 een dames- en herenmodezaak aan de Langenhorst in Rotterdam. Moulijn groeide op in de Bloklandstraat in het Oude Noorden van Rotterdam. Het muurtje waar hij in zijn jeugd eindeloos tegenaan had gevoetbald, was befaamd maar viel in de jaren zeventig ten prooi aan de stadsvernieuwing. In 1990 is elders in dezelfde straat een speelplaats voorzien van een nep-"muur van Coen Moulijn", ontworpen door de kunstenaar Hans Citroen.
