Es gehört geologisch zur Wildhorndecke und ist aus Kalken der Kreidezeit aufgebaut. Das Brienzer Rothorn wird von Brienz aus durch die Brienz-Rothorn-Bahn erschlossen, von Sörenberg führt eine Luftseilbahn fast zum Gipfel. Im Winter ist dies der Startpunkt zum Skigebiet beim Eisee. Dieses wird durch eine lange Galerie erreicht, welche einen wintersicheren Zugang ermöglicht. Bei der Bergstation der Luftseilbahn gibt es ein Restaurant. Etwa 100 Meter unterhalb der Luftseilbahn liegt das Berggasthaus Rothorn Kulm. Neben Zweier- und Achtbettzimmern bietet es auch ein Massenlager für die Übernachtung. Wanderer erreichen das Brienzer Rothorn zumeist von Süden (Brienz) oder Osten (vom Brünig oder von Schönbüel her kommend). Die Aufstiege auf der Nordseite, von Sörenberg her, sind steil und weniger häufig besucht. Der lange, von Interlaken her zum Brienzer Rothorn ziehende, Brienzergrat wird selten begangen. Dank seiner abgesetzten Lage nördlich des Brienzersees hat man von seinem Gipfel eine hervorragende Aussicht in die Berner Hochalpen. Aufgrund der Nähe des Jagdbanngebiets Augstmatthorn findet sich auf dem Brienzer Rothorn eine grosse Zahl von Steinböcken.
