Von 1960 bis 1963 arbeitete sie mit Ray Charles; berühmt wurde das 1960 aufgenommene Duett Baby It’s Cold Outside. Mit Sonny Rollins trat sie 1963 in Japan und 1964 in England auf, später arbeitete sie auch mit dem Pianisten Mulgrew Miller. 1970 gründete sie das Platten-Label Bet-Car Records, bei dem in der Folgezeit ein Großteil ihrer Alben erschien. Ende der 1970er Jahre trat sie in der Carnegie Hall und beim Newport Jazz Festival auf und unternahm eine Tournee durch die USA und Europa. Neben ihrer eigenen musikalischen Karriere erwarb sich Carter einen Ruf als "Patentante des Jazz"; zu den von ihr entdeckten oder geförderten Talenten gehören u.a. John Hicks, Mulgrew Miller, Cyrus Chestnut, Mark Shim, Dave Holland, Stephen Scott und Kenny Washington. 1993 eröffnete sie die Veranstaltungsreihe Jazz Ahead, bei der sie eine Woche lang mit zwanzig jungen Jazzmusikern arbeitete. 1987 trat sie gemeinsam mit Carmen McRae auf (The Carmen McRae-Betty Carter Duets). Für das Album Look What I Got von 1988 erhielt sie einen Grammy Award als beste Sängerin. Droppin' Things (1990) und It's Not About the Melody (1992) wurden für einen Grammy nominiert. 1997 verlieh ihr US-Präsident Bill Clinton die National Medal of Arts.
